    is this a must to have a DLR to communicate between two VMs located on SAME ESXI but in Different SUBNET ?

    Yes, since your virtual machines are on different subnets.

    Depends on the use-case and you could solve this problem in many different ways -

    1> Just use good old vlan-backed port-groups and route on an external device or virtual router of choice

    2> Bank on the NSX capabilities in terms of workload mobility across Layer-3 boundaries by using vxlan backed PG's and hook'em up to DLR or ESG (latter won't give you distributed capabilities outside your host

    So since this is an NSX forum , I'd bet the answer is "yes" , you want so solve your question by using DLR -
