    Can anyone help me, I want to authorize incoming user with roles defined in 'Manage Roles' screem for an api/service. let me know, how to do that.

    A user coming into the system would need to authenticate, once authenticated they would have any roles you granted their user.

    Do you mean to authorize a user associated with a request? If so this is not possible. The "Manage Roles" feature ONLY applies to identities accessing the Gateway with the Policy Manager or Restman. Message based authorization must be handled otherwise, such as group membership in an identity provider, SiteMinder, or XACML.
